肺癌是最常见的恶性肿瘤之一，预计到2025年，中国将成为肺癌患病率第一大国[1]。近年来，中药在肿瘤临床治疗中的作用引起了医学界的瞩目，合理开发和运用中医药的抗肿瘤作用成为研究的热点。
益气养阴散结方是湖北中医学院成肇智教授治疗肺癌的常用验方，为了进一步揭示其抗肿瘤的具体机制，本实验采用血清药理学方法，从细胞水平探讨益气养阴散节方对人肺癌A549细胞增殖及凋亡的影响。
1 资料与方法
1．1 细胞与细胞培养 人肺癌A549细胞由武汉大学生命科学院惠赠。常规培养、消化、传代。
1．2 含药血清的制备 益气养阴散结方由生黄芪、南沙参、莪术等组成，均购自湖北中医学院专家门诊部，传统方法煎制，浓缩为1 mg/ml。大、中、小剂量组分别按体质量以20 mg/kg、10 mg/kg、5 mg/kg给大鼠灌胃，2次/d，连续3 d，于末次给药后1 h，经右心室取血，通过离心取得含药血清[2]，置于-20℃保存备用。
1．3 方法
1．3．1 MTT比色法 将细胞接种于96孔培养板中，每孔的细胞浓度为1×105/ml，待细胞长满单层后，分别加入大、中、小剂量含药血清及对照组血清(不含药的大鼠血清)。每组6孔，培养48 h后，加入5 mg/ml的MTT20 μl，培养4 h。每孔加入DMSO200 μl，混匀，震荡培养板10 min。对照组调零，用酶标仪测各孔490nm波长光吸收值(OD值)，根据OD值判定含药血清对细胞增殖的影响。
1．3．2 Annexin V/PI法检测细胞凋亡 将细胞接种于24孔培养板中，每孔的细胞浓度为1×105/ml，待细胞长满单层后，加入中剂量含药血清及对照组血清，培养48 h后，用0．25%胰蛋白酶消化收集细胞，按照Annexin V/PI凋亡检测试剂盒说明操作，上流式细胞仪检测。
1．6 统计学方法
均用 SPSS12软件处理。

2．1 MTT法检测结果 药物组各组对细胞增殖的抑制作用均明显高于对照组(P

3 讨论
益气养阴散结方是由生黄芪、南沙参、莪术等组成，方中莪术行气破血消积，生黄芪补气升阳，调节机体免疫功能，同时又可防莪术耗气伤气之弊，沙参润肺滋阴，沙参之寒与黄芪之温相配，则寒而无伤脾之虞，燥而无劫阴之虑。诸药配伍，攻补兼施，共奏益气消瘀、扶正祛邪之功。实验证明，由以上药物所组成的方剂在临床肺癌的治疗中取得了肯定的效果[3-4]。
凋亡(apoptosis),也即程序性细胞死亡，是由基因控制的细胞的自我消亡。近年来的研究发现，正常机体内，细胞增殖与凋亡处在动态的平衡中，增殖过度与凋亡不足都是肿瘤发生的重要原因。因此通过阻断细胞增殖和诱导细胞凋亡都可以达到阻断肿瘤进展和改善预后的目的。近年来的研究显示，中药抗肿瘤作用的重要机制之一就是抑制细胞增殖和诱导细胞凋亡[5]。
本实验结果显示：与对照组相比，益气养阴散结方能明显抑制肺癌细胞的增殖，同时能明显诱导细胞凋亡，虽然其具体的机理还有待研究，但中药在抗肿瘤方面的广阔前景无疑是令人期待的。
