摘要:RNA干扰(RNAi)是一种转录后的基因缄默.它能触发某种转录后监控程序,导致特定mRNA单链的降解[1-3].我们利用RNAi技术,针对survivin基因的2个不同部位设计2对编码小干扰RNA(siRNA)的DNA模板,用基因重组的方法将其置于质粒pGCsilencer的U6启动子下,构建pGCsi-lencer-survivin siRNA重组质粒,转导人白血病K562细胞,以阻抑survivin基因的过度表达,诱导K562细胞凋亡.期望为肿瘤的基因治疗提供实验基础.
