摘要:目的:探索Snail基因与食管癌细胞侵袭迁移能力的关系。方法:体外合成Snail序列特异性双链小干扰RNA,转染人食管癌细胞EC9706,应用RT—PCR及Western blotting方法检测干扰效果,细胞划痕实验检测细胞迁移能力变化,Transwell小室侵袭实验检测细胞侵袭能力变化。结果:经Snail特异性siRNA干扰后细胞Snail基因及蛋白表达水平明显降低,细胞的侵袭迁移能力随之降低,差异均有统计学意义(均P〈0.05)。结论:Snail基因与食管癌细胞的侵袭迁移能力有关,采用siRNA干扰Snail基因的表达能有效逆转食管癌细胞的侵袭迁移能力。
