摘要:目的比较聚丙烯酰胺凝胶-银染(简称银染)、SYBR Green Ⅰ及溴化已锭(EB)染色方法在定量聚合酶链反应(Q-PCR)检测DNA的敏感性.方法分别用聚丙烯酰胺凝胶-银染、琼脂糖电泳-SYBR GreenⅠ染色及EB染色方法检测相同条件下PCR产物,进行光密度定量分析及比较.结果24个循环PCR检测DNA产物:SYBR Green Ⅰ染色与银染均见清晰带并可定量分析,而EB染色在28个循环次数尚可清晰带型;SYBR Green Ⅰ及EB染色历时30min,而银染则历时4h~5h.结论在检测DNA时,SYBR Green Ⅰ染色敏感性同银染,但比EB强;而且比银染更简单、省时、经济.
