摘要:RNA干扰(RNAi)是由双链RNA(dsRNA)引发的转录后基因沉默(PTGS)机制,具有普遍的生物学意义.dsRNA经DICER酶切割为21~23核苷酸长的小分子干扰RN段(siRNA),可以高效、特异地阻断体内特定基因表达,促使mRNA降解,诱使细胞表现出特定基因缺失的表型.由于能够迅速而简单地制备某个基因功能缺失表型,RNAi实验技术的发展将为基因或蛋白功能研究、药物筛选、信号通路分析、基因治疗等提供新的重要研究手段和技术平台.
