摘要:目的:肝脏内有两种纤维母细胞,一种称之为肝星形细胞(hepatic stellate cell HSC),另一种是位于门脉周围的纤维母细胞。对这两种细胞在形成肝纤维化(hepatic fibrosis HF)后的细胞形态采用免疫组化方法进行组织学鉴别。方法:Wistar大鼠48只,雄性,体质量(160~210)g。随机选取24只,分为4组,每组6只,使用30%CCl4/mineral oil(1m L/kg)行腹腔内注射,并且分别于注射1d,2d,3d,4d后麻醉处死,取新鲜肝脏。其余24只,同样随机分为4组,每组6只,分别行胆总管结扎术1d,2d,4d,7d后麻醉处死,取新鲜肝脏。用SABC法来进行检测,一抗分别为Vinculin及GFAP。结果:CCl4腹腔注射后2d中央静脉周围的HSCs在Vinculin和GFAP染色中呈相同的强阳性表达;BDL处置4d后门脉区纤维母细胞在Vinculin染色中显示阳性结果,而在GFAP染色中呈现阴性结果。结论:Vinculin在HSCs和门脉区域的纤维母细胞阳性表达结果相同,GFAP可对有活性的HSCs和门脉区域的纤维母细胞进行检测鉴别。
