摘要:目的探讨Notch信号通路在血管紧张素-Ⅱ(AngⅡ)诱导炎症反应中的作用。方法 THP-1细胞诱导分化为巨噬细胞,给予Ang II处理,用q RT-PCR和免疫印迹检测Notch1、Dll1、TLR-4、NFκB及炎性因子表达。采用si RNA沉默TLR-4或SN50阻断NFκB,观察炎性因子变化。结果巨噬细胞中Notch1、Dll1蛋白表达在Ang II处理后呈时间依赖性增加(P〈0.05)。TLR-4、NFκB、IL-1β和TNF-α表达在Ang II处理24 h后明显升高(P〈0.05),而IL-6表达明显降低(P〈0.05);TLR4沉默或NFκB阻断后,IL-1β、IL-6和TNF-α表达无明显变化。结论 Notch信号通路在Ang II诱导炎症反应中具有重要作用,针对Notch信号通路的调节可能有助于改善动脉粥样硬化。
